New Study Shows How Histamine Might Play a Role in Depression

A new study from Imperial College London and the University of South Carolina links high histamine to depression. The findings confirm prior studies that inflammation, which triggers histamine release, affects the hormone serotonin. Low levels of this crucial molecule in the brain seem to link to depression, according to this science.

If scientists can reproduce the study in humans, these findings could lead to new treatments for depression. Depression affects nearly 300 million people worldwide, according to the WHO, although it is likely a conservative estimate. Depression remains the leading cause of disability globally and dramatically contributes to the global burden of disease. It’s believed that this complex mental disorder stems from a combination of social, biological, and psychological factors.

Scientists have attempted to understand the root cause of depression for decades. Now, they reveal how histamine contributes to this devastating mental disorder. The body releases histamine as an immune response to a normally harmless foreign substance, such as pollen. When this happens, immune cells attack the perceived threat, and blood flow increases in affected areas.

This immune response allows the body to fight infections, but it can become compromised if histamine remains elevated. Chronic inflammation keeps histamine levels high, which weakens the immune system. Many health conditions cause inflammation, including cancer, diabetes, obesity, allergies, and even stress. The study, which appeared in The Journal of Neuroscience, reveals that inflammation-induced histamine might also contribute to depression.

Lead author Dr. Parastoo Hashemi, from Imperial’s Department of Bioengineering, said: “Inflammation could play a huge role in depression, and there is already strong evidence that patients with both depression and severe inflammation are the ones most likely not to respond to antidepressants.

“Our work shines a spotlight on histamine as a potential key player in depression. This, and its interactions with the ‘feel-good molecule’ serotonin, may thus be a crucial new avenue in improving serotonin-based treatments for depression.”

Many antidepressants target serotonin, commonly called the “feel-good hormone.” Most selective serotonin reuptake inhibitors (SSRIs) impede the re-absorption of serotonin in the brain. This allows the serotonin to circulate for a more extended period, enhancing mood and warding off depression.

SSRIs undoubtedly improve many lives, but an increasing number of people have become resistant to their effects. Experts believe that specific interactions between neurotransmitters, including serotonin and histamine, play a considerable role in treatment-resistant depression.

So, researchers focused their investigation on the complex relationship between histamine, serotonin, and SSRIs. For the study, they placed microelectrodes in the hippocampus of the brains of live mice to measure serotonin levels. The hippocampus helps regulate mood and plays a vital role in storing and recalling memories.

This method, also called fast-scan cyclic voltammetry (FSCV), allowed them to measure serotonin levels without harming the brain. The microelectrodes measure only five micrometers wide and do not endanger living tissue. After placing the microelectrodes, they gave half the mice lipopolysaccharide (LPS) injections, a toxin found in some bacteria that triggers inflammation. Scientists gave the other half, the control group, a saline solution.

The observations

The researchers observed that brain serotonin levels decreased after a few minutes of the LPS injection. In the control mice, serotonin levels did not change. This shows how quickly the brain responds to inflammation, which affects serotonin levels. Since LPS cannot cross the blood-brain barrier, it couldn’t have been responsible for causing the decrease in serotonin.

Upon investigating further, the researchers found that the inflammation triggered the release of histamine, which directly inhibited serotonin. This occurred when the histamine attached to inhibitory receptors on the serotonin neurons. Scientists believe a human study would reproduce this effect since humans also have these receptors on their serotonin neurons.

To see how SSRIs would affect histamine and serotonin, researchers administered them to the mice. Surprisingly, the control mice had higher serotonin levels than the other group. Researchers believe this occurred because the SSRIs caused histamine levels to rise, which canceled the serotonin boost.

Next, researchers gave the mice antihistamines to counter histamine’s inhibitory effects. This caused serotonin levels to rise back to normal levels. The findings confirm the scientists’ theory that histamine directly inhibits serotonin in the brain. The histamine-reducing drugs triggered a whole-body drop in histamine, unlike regular antihistamines taken for allergies, which only affect neurons.

Dr. Hashemi added: “Inflammation is a whole-body response and is therefore hugely complex. Depression is similarly complex, and the chemicals involved are affected in myriad ways by both genetic and environmental factors. Thus we need to look at more complex models of depression behaviors in both mice and humans to get a fuller picture of both histamine and serotonin’s roles in depression.”

According to Selin Malkoc, co-author of the study and associate professor of marketing at Ohio State University’s Fisher College of Business, they also had worse mental health outcomes.

“There is plenty of research which suggests that leisure has mental health benefits and that it can make us more productive and less stressed,” Malkoc said.

“But we find that if people start to believe that leisure is wasteful, they may end up being more depressed and more stressed.”

Thinking of leisure as wasteful increases stress

One study included 199 college students who rated their enjoyment of several leisure activities. They also completed questionnaires that measured their happiness, depression, anxiety, and stress levels.

Researchers asked them how much they agreed with five statements assessing their feelings about leisure time. For example, one statement said: “Time spent on leisure activities is often wasted time.”

Results found that the more participants agreed with leisure time being wasteful, the less they enjoyed leisurely activities. This held whether they engaged in active (exercising), passive (watching TV), or solitary (meditating) leisure activities.

What’s more, those who most strongly agreed with leisure time being unproductive felt the least happy. They also suffered from higher levels of depression, anxiety, and stress.

But, having fun with a larger goal in mind improves mental health

Another study involved 302 online volunteers who answered questions about how they celebrated Halloween in 2019. They could choose from activities such as going to a party or taking their kids trick-or-treating. Participants then rated how much they enjoyed their Halloween experience.

The results that prove the importance of leisure time

Results found that those who viewed leisure time as wasteful reported having less fun at events like parties.

“But those who participated in fun activities that fulfilled responsibilities, like trick or treating with your kids, didn’t see such a reduction in how much they enjoyed their Halloween,” said study co-author Gabriela Tonietto, an assistant professor of marketing at the Rutgers Business School.

Obsession with productivity doesn’t just affect the United States.

You might think that only overworked Americans shame leisure time, but it seems other countries share this belief. One study compared people in the U.S., India, and France. Of course, fewer people in France believed leisure was wasteful compared to the United States and India. However, those in France who looked down upon leisure still experienced the same adverse effects.

“We live in a global society, and there are people everywhere that hear the same messages about how important it is to be busy and productive,” Reczek said.

“And once you believe that and internalize the message that leisure is a waste, our results suggest you’re going to be more depressed and less happy, no matter where you live.”

It also surprised researchers that the disdain of leisure time affected the enjoyment of anything fun. It didn’t matter how long the leisure activity lasted or what people participated in; the results remained the same.

Skeptics of leisure time hold onto their beliefs.

In another study, researchers asked college students to watch a short funny cat video in the middle of an experiment. Some participants read articles beforehand that explained how leisure time could reduce stress and boost energy. Even still, the adverse effects of prioritizing productivity remained.

“These are students who are coming into the lab to answer surveys, which can be boring. In the middle of that, we give them a funny video to watch, which you would expect would be a nice break – and even then, some participants didn’t enjoy it as much,” Malkoc said.

“They had no way to use the time more productively. We were giving them a break from other, more boring activities. And still, those who believe leisure is wasteful didn’t think watching the videos was as fun as others did.”

As my interest in art grew and my ability developed, I realized that if I could learn to do this, I could learn to do anything. I started writing a novel at the age of 20 and spent the next few years badgering people to read it and correct the spelling. I know two things about myself that are true: I can’t paint when I’m drunk, and I’m terrible at spelling, but thanks to the creative medium of art, I am not afraid to fail or succeed.”

European Study Explains How Climate Change Threatens Freshwater Fish

Rising ocean temperatures caused by global warming may impact thousands of freshwater fish species. A European study found that a 1.5-degree Celsius rise in global average temperature would threaten about 3% of freshwater species. A 3.2-degree Celsius increase in temperature would impact over 50% of the habitat for one-third of freshwater fish. Scientists say that two factors pose the greatest threat to fish: extremes in water temperature and streamflow.

Radboud University led the study, collaborating with Utrecht University, PBL Netherlands Environmental Assessment Agency, Leiden University, and others. It was published in Nature Communications on March 15th, 2021.

Previous studies have already investigated the possible implications of climate change on various plant and animal species.

freshwater fish“However, freshwater fish species have been largely ignored, even though they represent approximately a quarter of the global known vertebrate diversity,” says Valerio Barbarossa, lead author of the paper. The study is the first to assess the possible impact of global warming on around 11,500 freshwater fish species globally.

Scientists say that the impacts largely depend on future greenhouse gas emission levels. According to the EPA, unmitigated greenhouse gas emissions would result in a 62% decline in freshwater fish habitats by 2100. However, this number lowers drastically to only 12% if we work to reduce emissions.

A mitigation strategy would preserve coldwater habitats throughout the Mountain West and Appalachia regions. It would also avoid an estimated $380 million to $1.5 billion in total recreational fishing losses through 2100.  Furthermore, around 440,000 acres of stream habitat would disappear if we don’t take action against climate change. This reduces drastically to only 85,000 acres of lost habitat for freshwater fish under a mitigation scenario.

The EPA report says that less economically valuable fisheries will replace many coldwater species without global GHG mitigation. Unabated climate change would affect the Mountain West and Appalachia regions the most. In fact, by not reducing emissions, coldwater fisheries would virtually disappear from Appalachia. It would also alter the stream habitat from warm to rough in Texas, Oklahoma, Kansas, and Florida.

Climate change will impact freshwater fish regardless of mitigation strategies.

No matter how much we limit greenhouse gas emissions, fish and other marine animals will still feel the effects. However, by reducing the damage as much as possible, we can preserve the habitats for most fish species. With a global rise of 3.2 degrees Celsius, over one-third of freshwater fish would lose around 50% of their habitats. This scenario would occur if governments stick to their pledges for 2030 but don’t commit to any further emission cuts.

Limiting global warming to 2 degrees would threaten half the habitat for only 9% of freshwater fish species. Further limits to 1.5 degrees would reduce the number of species at risk to just 4%.

“These numbers indicate that limiting global warming really matters for freshwater fish species, just as previous research has shown that it matters for species in terrestrial systems,” says Barbarossa.

How water temperature and flow changes will affect freshwater fish

The researchers created models to see how extremes in water flow and temperature would impact freshwater fish. They also identified where the greatest extremes might occur in future scenarios.

“Water temperature and water flow are two key habitat factors for freshwater fish species. Climate change will amplify extremes in flow and temperature, which may reduce the amount of suitable habitat. This, in turn, is an important indicator of extinction risk,” says Aafke Schipper, environmental researcher at Radboud University and PBL and co-author of the study.

The team found that water temperature changes would pose more of a threat than flow extremes. This indicates that global warming will cause rising water temperatures across the world. However, the research revealed that freshwater fish in tropical waters would face the highest threat due to extremely warm or tropical waters.

“The numbers of species at risk represent a worst-case scenario in the sense that we assume that fish will not be able to move to other parts of the watershed or adapt to changed conditions,” continues Barbarossa. “We have also considered a scenario in which species could move freely across the watershed and “escape” altered conditions. In that case, climate change threats would be substantially lower. However, many freshwater systems are fragmented, which impedes fish from moving to more suitable conditions.”

Many rivers around the world now have one or more human-made barriers like dams, culverts, sluices, or weirs. While these help with flood control, they also impede fish from migrating due to reduced connectivity of habitats. As the globe continues to warm, this poses a huge threat to freshwater fish populations. The authors of the study say that this increases the urgency to limit global warming to ensure freshwater fish biodiversity.

Global warming also causes the oceans to become more acidic due to an increase in hydrogen ions. This chemical reaction results in changes in the water’s pH, and calcium carbonate saturation levels decrease. Because of the huge influx of carbon dioxide, the ocean can’t hold as much oxygen. The combination of all these changes may result in mass extinctions, or at the very least, less healthy marine life.

Asthma researchers have identified a new severe asthma species, showing air pollution as a likely contributor. Asthma affects over 300 million people worldwide. The most severe strain, known as non-Th2, or non-atopic childhood asthma, comprises most cases.

Hyunok Choi, an associate professor at the Lehigh University College of Health, says 85% of people have this type of asthma.  Low-income countries are disproportionately affected by this strain. However, asthma researchers still don’t know if non-Th2 is a distinct disease or just a set of symptoms. The purpose of this study was to find out more about this strain of asthma.

Says Choi:

“Non-Th2 asthma is associated with very poor prognosis in children and great, life-long suffering due to the absence of effective therapies. There is an urgent need to better understand its mechanistic origin to enable early diagnosis and to stop the progression of the disease before it becomes severe.”

Studies show that around 50% of children with poorly managed asthma will have severe adult cases. Currently, many doctors follow a near-universal protocol when it comes to treating asthma. However, Choi says this approach doesn’t work with all patients and contributes to further strain on the economy.

“The primary reason for lack of therapeutic and preventive measures is that no etiologic, or causal, driver has ever been identified for the non-Th2 asthma,” says Choi.

The groundbreaking epidemiological study led by Choi shows that non-Th2 is, in fact, a distinct disease. It also found that early childhood exposure to air pollution, particularly Benzo[a]pyrene, likely causes the development of this asthma strain. Asthma researchers have categorized asthma by two major groups of symptoms: T helper cell high (Th2-high) and T helper cell low (non-Th2). Th2-high includes early childhood allergies to common pollutants like pet dander, tree pollens, or mold. On the other hand, non-TH2 doesn’t occur due to an allergic response.

Asthma researchers have much less understanding of non-TH2 asthma, which is more severe and harder to treat. However, the new findings from this study may help lessen the severity of the disease. Choi explains:

“The identification of non-Th2 asthma as a distinct disease, with early exposure to Benzo[a]pyrene as a driver, has the potential to impact tens of millions of sufferers, since this would make it possible to intervene before the onset of irreversible respiratory injuries.”

The asthma researchers tested two similar groups of children from two different towns in the Czech Republic for the study. One group of children was from an industrial city called Ostrava. The other group lived in a relatively rural area of Southern Bohemia. 385 children participated in the study: 194 with asthma and 191 in the control group.

According to the study, Ostrava is a heavily industrial area with prevalent coal mining, coal processing, and metallurgical refinement. During their investigation, researchers discovered highly elevated levels of Benzo[a]pyrene. In fact, the district-level ambient mean measured 11 times higher than the recommended outdoor and indoor levels.

Asthma researchers found a direct link between this chemical and the development of non-Th2 asthma. They also discovered that exposure to the fossil fuel byproduct led to depressed systemic oxidant levels.

“Contrary to the current body of evidence supporting adult onset of non-atopic asthma, our data suggest for the first time that the lung function deficit and suppressed oxidative stress levels during early childhood are critical sentinel events preceding non-atopic asthma,” says Choi.
